Effect of Ethanol on Na-K-ATPase Activity of Rabbit Kidney


Abstract
In order to determine the effect of ethanol on Na-K-ATPase and to clarify the action mechanisms of ethanol, the effects of n-alcohols including ethanol were observed in rabbit kidney homogenate. Na-K-ATPase activity was inhibited by ethanol in higher concentrations than 1.0%, in dose-dependent manner, but Mg-ATPase activity was affected in only 10 and 15% concentrations.
The effects of ethanol on Na-K-ATPase activity were unaffected by preincubation time and the enzyme concentrations. For the n-aliphatic alcohols, there exists a linear relationship between the logarithm of equiactive concentrations and the number of carbon atms in the aliphatic chain.. Arrhenius plot showed two break points at 20¡Æ and 30¡ÆC in the control medium which decreased to 18¡Æ and 28¡ÆC, respectively, by 7% ethanol.
These results indicate that ethanol inhibited Na-K-ATPase activity by interacting with the hydrophobic region and resulting in a disturbance of microenvironment of lipids associated with the enzyme. In addition, the hydrophobic bonds are shown to have an important role in maintaining the Na-K-ATPase activity.
